When I started up Profiler today I noticed than I have no checkboxes in the collection list:

I've tried re-installing (latest beta, build 1606), and have also done a database repair. No luck.
Not sure, but this may have occured after the latest batch of Microsoft updates.

I'm pretty sure you can enable/disable them via the options menu.

Correct.
Menu would be "Collection -> Flags -> Show Checkboxes"
It all seems so stupid, it makes me want to give up!
But why should I give up, when it all seems so stupid?

On mine it is "Collection -> Flags -> Show Flag Indicators"

This is what happens if working with a localised version and being to lazy to switch to the original team up.

You are correct, of course.
It all seems so stupid, it makes me want to give up!
But why should I give up, when it all seems so stupid?

Thanks guys!

Not sure if it's a combination of keystrokes or what, but it's happened to me a few times before over the years.

It used to turn itself off much more frequently.  I've not noticed it happening in a while now.  Strange.
