There seems to be something wrong with your layout file. Open another layout (View --> Open Layout...), preferably the default one that comes with the setup files, close DVD Profiler and then start it again.

If the same message as before appears, you could probably try to reinstall DVD Profiler, so that the original layout files are restored.

EDIT: But remember to backup everything (databases, layout files etc.) before you uninstall the software!

Ha, silly me  . Choose "Diagnostic Startup" and "Use a default layout". If DVDP starts correctly, then the layout file that was loaded when you rebooted your PC is damaged.
Sorry, it's too late I guess. Or I seem to have lost my ability to read... 

If you have a recent backup, you can reinstall DVDP, like I suggested before. Hopefully that restores the currupted files.

An alternative would be to rename the layout file you were using (e.g. MyLayout.dpl --> MyLayout.dpl.bak). It's located in "C:\Documents and Settings\<USERNAME>\My Documents\DVD Profiler\Layouts" on WinXP or "C:\Users\<USERNAME>\Documents\DVD Profiler\Layouts" on Vista/7.
If that doesn't help and the error message still says "... stopping at: Setting up layout", then there seems to be more damage and you should reinstall DVDP anyway. Don't forget to backup the user files by making a copy of the whole folder "C:\Users\<USERNAME>\Documents\DVD Profiler" (or "C:\Users\<USERNAME>\Documents\DVD Profiler").

Another thing you could try would be to run a database repair (Start Menu --> DVD Profiler --> Database Repair Utility). Make a backup before, just to be sure. That doesn't fix anything related to layouts, but who knows, perhaps the error message is wrong

Do you have other layouts than the default layouts installed on DVDProfiler?
Or did you remove any manually lately?

EDIT: You may want to try the following:
Click on Start  -> Run
Type "regedit"
In the now opening window navigate to the key "HKEY_Current_User -> Software -> Invelos Software -> DVD Profiler".
Single left click on "DVD Profiler"
In the right part of the window now navigate to a subkey named "LayoutFileName" the value next to this subkey is supposed to mirror the location of your layout file (Documents -> DVD Profiler -> Layouts -> xyz.dpl)
If either the path or the filename are incorrect you can correct it here by single rightclick on "LayoutFileName" -> "Change" and correcting the value in the now opening window.
If in doubt point the value to a file named "Invelos Default.dpl"

I did this and it worked fine for a while, but now it has come back.

I havent made any changes to layhout or anything other than adding movies to the database
